Nintendo designer Shigeru Miyamoto stated that he wanted to create a location that he could use across multiple games of different genres. Wuhu Island is the culmination of this idea, and will go on to appear in other games in the future.
However, the island has already shown up before. In one of the early Wii demos, which may have inspired this game's Island Flyover, a very similar island appeared. The jogging sections of Wii Fit also appear to be set on Wuhu Island.
Going back further, the island shares many of its landmarks with both Holiday Island and Crescent Island from Pilotwings 64.
The name is likely a reference to Mario's joyful cries of "Woohoo".
The name of the island's luxury oceanliner is "Queen Peach", a reference to Princess Peach of the Mario series.
In the Island Flyover, when you fly close to the houses of Hill Village, you can hear the Mario "Miss" tune from Super Mario Bros.
At night, the Information icon for the village says: "It sounds like someone is playing an NES. Sounds like he needs help though."
The Power Cruising game is essentially a jetski race in the style of the Wave Race series. In fact, the game's music seems to be inspired heavily by the series.
Also, the sound effect when you start any of the races appears to have been taken directly from Mario Kart 64.

In the U.S. version of the game only, one of the stickers you can earn in Archery is named "It's a secret to everybody".
This phrase was first uttered by characters in secret caves in the original The Legend of Zelda (NES 1986).
